A novel combination of elevated hydrostatic pressure and solubilizing chemicals was developed for two key sample preparation techniques: enzymatic protein digestion and cell lysis. This method allowed us to enhance the depth of proteomic profiling of cell lysates at least 2-fold while increasing the throughput of sample preparation greater than 20-fold when compared to the conventional method and producing a nearly 3-fold increase in the number of putative membrane proteins.
