Microflow DIA Using 15 min Gradients Analyzes 40 Tumor Proteomes per Day and Effectively Detects Promising Protein Biomarkers

The typical gradients used with nanoflow chromatography are 2-3 hours, allowing only 8 complex proteomes. Increasingly microflow chromatography has been combined with SWATH Acquisition MS to accelerate quantitative proteomics. Very good protein quantitation numbers have been demonstrated with shortened gradients. In this study, we explore whether using a 15 min microflow LC gradientwith SWATH MS, enabling throughput of 40 proteome samples per day, is robust, practical and effective for prostate cancer tissue biomarker discovery (Figure 1).
